چکیده

Background Signalling by NO/cGMP is very important for regulation of vascular tone and gastrointestinal motility. One important signalling pathway of cGMP-dependent protein kinase type I (cGKI) is mediated by IRAG (IP3 Receptor Associated cGKI substrate) which is highly expressed in smooth muscle of cardiovascular and gastrointestinal tissue. In order to elucidate the physiological role of IRAG we generated IRAG knockout mice by targeted deletion of exon 3. These mice show an enlarged gastrointestinal tract including pylorus stenosis.

IRAG and novel PKG targeting in the cardiovascular system.

Signaling by nitric oxide (NO) determines several cardiovascular functions including blood pressure regulation, cardiac and smooth muscle hypertrophy, and platelet function. NO stimulates the synthesis of cGMP by soluble guanylyl cyclases and thereby activates cGMP-dependent protein kinases (PKGs), mediating most of the cGMP functions. Hence, an elucidation of the PKG signaling cascade is essen...

IRAG determines nitric oxide- and atrial natriuretic peptide-mediated smooth muscle relaxation.

AIMS Nitric oxide (NO) and atrial natriuretic peptide (ANP) signalling via cGMP controls smooth muscle tone. One important signalling pathway of cGMP-dependent protein kinase type I (cGKI) is mediated by IRAG (IP(3) receptor associated cGKI substrate) which is highly expressed in smooth muscle tissues. To elucidate the role of IRAG for NO- and ANP-mediated smooth muscle tone regulation, cGKI lo...

The effect of protocol of weaning from mechanical ventilation on gastrointestinal function in people undergoing coronary artery bypass graft surgery (CABG)

Abstract Aim. The aim of this study was to examine the effect of protocol of weaning from mechanical ventilation on the gastrointestinal function in people undergoing CABG. Background. The most common problems after cardiac surgery in the intensive care unit are change in vital signs, respiratory problems, unsuccessful separation from mechanical ventilation, and gastrointestinal problems. Due t...

Function of IRAG and the phosphorylation of the InsP3R-I for the NO/cGMP-dependent inhibition of platelet aggregation

Results We investigated the relevance of IRAG and the cGKI stimulated phosphorylation of the calcium channel InsP3R-I for the NO/cGMP-dependent inhibition of platelet aggregation and adhesion. After incubation with different agonists (collagen, thrombin, TxA2) we performed aggregation experiments with platelets of WT and IRAG-KO mice, thereby the IRAG-KO platelets aggregated stronger than the W...

Functional dissection of the cGK substrate IRAG using transgenic models

NO/cGMP signalling via cGMP-dependent kinase I (cGKI) induces a variety of physiological functions comprising relaxation of smooth muscle and inhibition of platelet aggregation. Several signalling pathways of cGKI exist including the interaction of the cGKIβ isozyme with the inositol 1,4,5-trisphosphate receptor I (IP3RI) associated protein cGMP kinase substrate (IRAG). To get insight into the ...
